OECD GUIDELINE FOR THE TESTING OF CHEMICALS In Vitro Mammalian Chromosome Aberration Test

ثبت نشده
چکیده

1. The purpose of the in vitro chromosome aberration test is to identify agents that cause structural chromosome aberrations in cultured mammalian cells (1)(2)(3). Structural aberrations may be of two types, chromosome or chromatid. With the majority of chemical mutagens, induced aberrations are of the chromatid type, but chromosome-type aberrations also occur. An increase in polyploidy may indicate that a chemical has the potential to induce numerical aberrations. However, this guideline is not designed to measure numerical aberrations and is not routinely used for that purpose. Chromosome mutations and related events are the cause of many human genetic diseases and there is substantial evidence that chromosome mutations and related events causing alterations in oncogenes and tumour suppressor genes of somatic cells are involved in cancer induction in humans and experimental animals.

OECD GUIDELINE FOR THE TESTING OF CHEMICALS PROPOSAL FOR UPDATING TEST GUIDELINE 473 In Vitro Mammalian Chromosomal Aberration Test

1. The OECD Guidelines for the Testing of Chemicals are periodically reviewed in the light of scientific progress, changing regulatory needs, and animal welfare considerations. The original Test Guideline 473 was adopted in 1983. In 1997 a revised version was issued, based on scientific progress made to that date. Modifications within the current version reflect nearly thirty years of experienc...

Oecd Guideline for the Testing of Chemicals

1. OECD guidelines are available for a wide range of in vitro mutation assays that are able to detect chromosomal and/or gene mutations. However, while in vivo assays in somatic tissues are desirable components of a thorough genetic toxicity testing programme, aside from the little-used (Wahnschaffe et al., 2005) rodent Spot Test (TG 484), OECD guidelines are available only for assays capable o...

DRAFT OECD GUIDELINE FOR THE TESTING OF CHEMICALS Rodent alkaline single cell gel electrophoresis (Comet) assay INTRODUCTION

1. OECD Test Guidelines (TGs) are available for a wide range of in vitro genotoxicity assays that are able to detect DNA damage, gene mutation and/or chromosomal aberrations. There are TGs for in vivo endpoints (i.e. chromosomal aberrations, gene mutations and DNA repair as unscheduled DNA synthesis); however, these do not directly measure DNA damage. The alkaline single cell gel electrophoresi...

Analysis of published data for top concentration considerations in mammalian cell genotoxicity testing.

The ability of the in vitro mammalian cell tests currently used to identify genotoxins has been shown to be limited by a high rate of false-positive results, triggering further unnecessary testing in vivo. During an European Centre for the Validation of Alternative Methods workshop on how to improve the specificity of these assays, testing at high concentrations was identified as one possible s...

Mutagenicity testing with transgenic mice. Part II: Comparison with the mouse spot test

The mouse spot test, an in vivo mutation assay, has been used to assess a number of chemicals. It is at present the only in vivo mammalian test system capable of detecting somatic gene mutations according to OECD guidelines (OECD guideline 484). It is however rather insensitive, animal consuming and expensive type of test. More recently several assays using transgenic animals have been develope...
